the shorter balls up over the net with fewer going under. It's harder to
get the first flat serves in at least at the beginning. I felt better
placement making and improved plow through on my single hand backhand.
Maybe it was just that the new strings work better. I tended to leave the
poly mains too long until the cross multifilament broke, when the mains were
long time over due. |

俺先后用过两个proofreading，先是用NEB的Deep Vent,这个酶的忠实度狠好，俺用来构
建质粒，35cycles,将近1k的三个片段拿去测序，没有错误。
后来作RT PCR的时候因为template是total poly A RNA，而Deep Vent没有Hot start，结
果不是太consistant，有时候有，有时候没有，产率也偏低。俺就换到Pfx plantium
(Invitrogen)，号称市面上忠实度最高的，现在所有RT PCR都作出来了，35cycles,3K的
片段。先拿了一个基因的三个克隆测序，结果是在两端大致1.5k的长度上DNA序列有2-4个
错误，氨基酸序列没有错误。而且由于俺同时测了三个克隆，以后可以用没有错误的克隆
拼接成error free的一个全长片段。
总的来说，Mg的浓度不要太高，酶的用量要尽可能少，然后就是wait and pray了，呵和
。

狭义上的polyA signal，指的是RNA序列上的一个Conserved motif: AATAAA（AAUAAA）
。CPSF蛋白识别并结合该位点，在CstF的辅助下切割下游pre-mRNA，再由PAP完成加尾
。当然整个过程还需要其他一系列蛋白因子的共同左右，不细说了。
你的问题涉及到CstF这个关键因子，它识别并结合AATAAA下游一段GU rich region，对
于有效的RNA Polyadenylation processing是不可或缺的，尽管这段序列不像poly
signal motif那样具有比较严格的consensus sequence。而在你的第二个版本中，并没
有包含这段序列。 |

2-vinylpyridine) is coordinated with the copper salt. I did the synthesis
based on literature, which says the solution is colorless after passing
through alumina column. Maybe there is a tricky issue that I don't realize.

know the tricks... since my polymer also can form ligation with copper and
after twice the column and it is still light green. However, isopropyl
alcohol definitely cannot get any ligands out, at least for my polymer.
