然后用FPG这种能识别8-oxoG的酶来切,被切的基因组位点会产生nick,阻碍PCR
polymerase,基于这样的原理做qPCR定量来研究某段序列对于oxidative damage的反
应。

如果用高浓度短时间刺激 (看文献是500uM 30min)之后用PBS洗掉,这个oxidation
的状态能维持多久?
我是希望这个oxidation状态能维持时间长一些,然后看我的酶分子是不是被氧化了?
并且这种氧化是不是改变了酶底物的状态和多少。所以希望酶的氧化状态能保持久一些
,从而容易观察到和对照的区别。
