星期开始的抗议活动采用“Ni Macron Ni Le Pen”（即两者都不要）这个口号至少代
表了一半选民的心声。抗议演变成暴动正是幕后一些人梦寐以求的。
大部分选民心里都清楚，按正常情形，目前勒庞以合法途径夺取政权的可能性是微乎其
微的。距离第二轮投票还有十天，没有意外的话，马克隆躺着也能进爱丽舍宫。
但是大选形势瞬息万变，去年美国大选投票前，很多人看好希拉里（躺着也能进白宫）
，突然冒出科米的一封信，打乱了政治精英们的计划。
当然不能把勒庞与川普相比，两者相差甚远。第一轮投票结果揭晓后，勒庞喜形于色的
表情与川普获胜后一脸严肃形成鲜明的对比。只得了百分之二十几的选民支持，勒庞竟
然能笑得出来。
尽管目前的“Ni Macron Ni Le Pen”抗议活动对勒庞选情来说有利无害，但是可以肯
定，这个口号不是她能想出来的，这个活动也不是她能策划的，她缺乏这样的政治谋略。
唯一可以让勒庞反败为胜的可能性就是今后的十天里“Ni Macron Ni Le Pen”抗议演

键性的实验支持。搜了一下通讯作者的背景，好像也没有抱过什么大腿，他自己也没什
么呼风唤雨的能力，这样的工作居然也能发NI？是审稿人的脑子秀逗了还是我才疏学浅
看不出这文章的高深之处？

我觉得这篇文章烂的原因：
1 题目不够准确。Autophagy proteins的缺失影响了Inflamamsome的功能，这是文章里
面用两种敲除老鼠证实的，但是不能说 Autophagy proteins regulate innate immune
responses。另外就是 the release of mitochondrial DNA mediated by the NALP3
inflammasome，这个结论的实验证据太单薄了，只有Figure 6d这一个实验证据，觉着
这么核心的结论还是应该多角度证明一下的。

由于必须用带N端pelB信号肽的pET26载体,并且N端靠近活性中心,所以只能把His-Tag放
C端,而且我已经试了放N端,蛋白彻底消失,估计N端Tag导致不稳定,降解了.
那么,除了Ni柱以外还有没有别的比较实用的办法纯化这个蛋白呢?我要用它作酶活鉴定
,所以需要纯化分离.蛋白大小在46kD.理论pI6.9. |

再复性.我是利用Triton X-100可以洗脱SDS的特点利用SDS变性,然后Triton X100洗脱
复性,然后过柱. 肯定不会对所有蛋白适用,但是起码我遇到的都work (包括一个>100
Kd的酶).
我是用的sonication water bath, 也可以用probe sonication再加SDS,都OK,但是注意
dilution的比例和体积.

发帖数: 330 | 24 Ni跟Co氧化都很快， Ni可能稍好一点。但这两东东在Po2>10E-15atm 以上都会慢慢氧
化，尤其是表面。你可以先看看Ellingham 氧化图 （可能拼错了），大概估计一下。
至于NiO跟Co 氧化物有没有催化性能，这要看你具体想催化啥反应，有些反应是纯金属
催化的，有些反应氧化物的催化性能好。 |
