因为我的peptides上有磷酸根, 所以想试一下negative ion mode,
mobile phase
A: 100% H2O 0.1% NH4OH,
B: 5% H2O 95 ACN 0.1% NH4OH
自己pack的 C18 capillary column
在一开始就碰到问题,
发现capillary column很容易堵住, 压力狂升, 最后系统shut down
有时候还能在tip上发现一些白色的crystal, 不知道啥东西
希望版上有经验的同学 指点指点 |

use any TEA in the previous wash.
The ion exchange (IX) solid phase (SP) contains a linker-piperazine with pKa
of 6, so I load the orotic acid (ORA, pKa1=2.8) with a pH 4.4 (half of 2.8+
6) solution to make sure both SP and ORA are ionized. Then I use 2%Formic
Acid/water (pH=2) to wash (should not elute any ORA out), and then use NH4OH
in MeOH/ACN to elute ORA. But the result shows the acidic wash step elute a

我主要用piranha来洗gold晶体,但它洗 glassware 的效果那叫一个好,虽然很危险。
开始我们组里人还不大敢用,后来不管什么glassware都用piranha,一整一大桶,搞得
我头都大了。
还有一个清洗办法效果也不错,用来洗wafer和glass, 就是SC1, 是指半导体工业里的
standard cleaning 1。 30%H2O2 加 NH4OH 加 DI water = 1:1:3 或者 1:1:4。
80摄氏度煮45min到一小时。稍微麻烦点,不过安全多了。
我们煮silicon wafer的时候把一个teflon底座开一些槽,将wafer插进去。
有些glassware奇形怪状的不好加热,一个办法是用水浴,因为SC1在80度左右就沸了。
当然还有SC2,去除普通金属的效果较好,一般用不上。
