如果你很想了解confocal或者更高级的two-photon,下面两篇paper对你会有帮助。
1.Ojcius DM et al.Immunology and the confocal microscope. Res Immunol 1996
2.Cahalan MD et al.Two-photon tissue imaging: seeing the immune system in a
fresh light.Nat Rev Immunol 2002
ZL1999 (胖头猫)提到了confocal和一般fluoresent microscopy的区别,除了有一点我
不太认同,根据我的理解,confocal microscopy的photobleaching问题应该比一般的
fluoresent microscopy要小,因为前者illuminate smaller volume of your sample。
如果你要做的是培养的细胞,你可以不用改protocal,我一般用8-well chamber slide

问下版上大牛,我是学统计与阵列信号处理的,现在有一做fluorescence microscopy
的OFFEER,说是要我做microscopy的定位精度的理论分析和软件开发的工作。
感觉信号处理方面不太好出文章,怕不太匹配难业,另外做fluorescence microscopy
好不好找工作啊,谢谢

confocal用的是PMT detector (大部分),这个的对光的检测效率(quantum
efficiency)在30-40%左右,对red 或者far red的检测效率更低
普通的fluorescent microscopy 用的是CCD 或者EMCCCD,效率在60-70%
confocal由于pinhole的存在,不但屏蔽了out focus的光子,同时也屏蔽了很多有效的
光子。
这两个原因,造成了confocal比普通的fluoresent microscopy的bleaching严重

atomic force microscopy的方法以及在2D materials surface and interfaces
characterization的应用;也涉及一些scanning tunneling microscopy和surface
science。文章目前出了一半的样子,有Nature子刊和ACS Nano 和一些领域内IF 4左右

去看看Xiaowei Zhuang, Hell, Gustafsson的文章,基本全是Science
Nature,他们做出来的技术,转眼Leica, Nikon做成系统,一套百万以上。
不过这些人都是做物理出身的,发展新microscopy技术主要是物理和光学工程,到后来才要做生物的人。

对女生来说如果以后能找到学校里比如microscopy facility里的位置其实也不错
但见仁见智
如果独立做研究的话要找到比较好的有意思的课题
这个需要广泛阅读还有建立合作关系

我很同意你的观点。
交叉领域虽然一般都比较新,容易出热点,但是对于中小lab来说,难度还是太大。生
物物理这一块,就我不多的了解而言,水也很深。我虽然不懂super resolution
microscopy,但我以前的实验室做的也可以算有点沾边。我的PI就是物理/建模出身,
从小到大数学很强,编程也不错,人很聪明。phd时期做了建模,博士后时期做了一些
生物物理仪器的开发,同时进入了免疫等生物领域。博士后时一直有大老板罩着,成绩
很好。可是独立之后,生存不易。因为你光开发出fancy的仪器不够,没有一个合适的
生物问题,那些传统生物学家是不会鸟你的。可是你要申funding的话,生物类的比仪
器类的要好申太多了。

打个比方说,一百年前的microscopy,没有荧光标记,只靠样品本身的吸收反射来看结
构,分辨率再高,你还是说不清这些结构到底是啥构成的。引入了fluorescence
spectroscopy的概念以后,上了一个荧光标记,通过滤波,只看这一种荧光成份,得到
的信息就有了特异性,microscopy在生物上才成了必不可少的工具。这就是
spectroscopy给microscopy带来的革命。
如果想七八种荧光标记一起上,按纯microscopy的思路,就得不停的换滤镜,取七八张
图,要花七八倍的时间,对活细胞是很不可行的。但是如果你对样品能做快速的荧光全
谱测量,只要取一幅图,图上每点都是个全谱,然后通过谱分析分解七八种标记。这样
可以做同时七八种荧光的显微电影,然后观察每个成份之间动态的相对关系。这就是高
速的fluorescence emission spectroscopy 和microscopy结合,加大信息通量,提高
分析能力。
谢的raman,针对的是没用荧光的物质,通过做raman spectrcopy的信息,让本来没什

从根本上说,庄做的是microscopy,谢是spectroscopy主打结合microscopy。光能探测
到的信息,远不止位置颜色这么一点。raman,荧光寿命等等成像方式,都是要求在
microscopy的基础上,加上高灵敏高速单点spectrocopy,来得到更多的信息。谢的路
子,是提高每点的信息含量。超高分辨microscopy,走的是提高分辨率的路子,具体到
每一个图像点,他们看到的信息与传统microscopy并无分别。到底是超高分辨好,还是
分辨率不变提高信息含量好,这要看具体应用。
谢虽然比庄资格老,但是他的技术很长时间以来一直受激光技术的限制,他本人说这个
最近轰动的SRS,是他十几年前就想做的,但是直到近几年,才有人研制出能满足要求
的激光。相比较而言,庄的技术幸运的多。

庄小威,1972年生于江苏省如皋县,是一位华裔美籍生物物理学家,哈佛大学化学与化
学生物、物理学教授,创办有庄小威实验室。[1][2][3] 她也是霍华德·休斯医学研究
所的研究员。[4]
1987年,她毕业苏州中学的科大少年班预备班。1991年,她获得中国科学技术大学物理
学学士;1997年,获美国加州大学伯克利分校物理学博士。她在1997年至2001年于斯坦
福大学攻读博士后。[5] 从她在哈佛建立自己的实验室开始,她带领研究团队发展超分
辨率显微镜技术(英语:super resolution microscopy)[6],识别了个体病毒粒子进
入细胞的机理[7],并用单分子技术从本质上研究核酸与蛋白的相互作用。
庄博士利用自己开发的超灵敏的光学成像技术来监测单一生物分子和联合体在体外或活
细胞内的瞬时行为,主要集中在理解病毒如何进入细胞及核酸-蛋白质相互作用。她拍
摄到单一枚感冒病毒如何影响一枚细胞,是首次有科学家记录到这过程。
