向高分子量的区域，噪音少
PMF是peptide mass fingerprint，有了现在的tandem mass spec直接测序，好像PMF用

use. For example, trypsin cleaves c-terminus of K and R (not if the
following amino acid is P) so you could get peptides with various length.
Each mass spectrometer has its own mass range that will determine how big/
small it can detect. And if you use LC-MS/MS instrument, the analytical
column also contributes to the detection. C18 columns can't retain
hydrophilic peptides very well. |

还有，通过sds－page来判断蛋白质大小的这种方法太不准确了，有些蛋白（没有任何
posttranslational修饰）在page胶甚至可以表现出2倍于理论分子量的size，所以你不
能说这个33kd的蛋白质就一定不是你的target protein。
当然size上的不一致也有可能是蛋白的修饰引起的，尤其是在细胞里提取的蛋白

1， 两个facility基本都发现同一批蛋白，高级的机器果然比旧机器好，发现更多的蛋
白，而且发现的peptide数目要多很多很多。
2， 旧机器发现的冠军蛋白A，在新机器下面不再是冠军蛋白。
3，新的冠军蛋白三周前search还是37kDa, 上周NCBI居然update成了47kDa, N
terminal多出一串amino acid，所以基本符合sds-page胶的预测 (45kda)。

发帖数: 190 | 33 我有一未知化合物需要鉴定。Mass Spec中心先是做了个Fourier transform mass
spectrometry，打出来的分子量是173。07842。他们用3-hydroxypicolinic acid,
Caffeine,CHCA matrix作为标准。但我觉得他们的标准有问题。比如3-
hydroxypicolinic acid，我查到的分子量是139。11，与我计算得出的139。1088很一
致。但他们MS上打出来的M+是140。03421，与我的计算的结果相差0。07左右, 挺大的
。而他们认为140。03421只跟标准相差0。07个PPM。另外两个标准也与我计算的相差蛮
大。不知道MS机器是不是有他们自己分子量标准，和我们平常用的不太一样。
然后, 他们又做了个MS/MS。但不知道为什么，我个分子是173（M+），经过MS/MS过，
最主要的峰仍然是173。可能是他们用的能量不够。他们最后的结论是这个化合物在他
们的database没有。但我觉得这是个比较常见的化合物。
最后附上他们做的两个MS图，请做MS大牛帮我看看这到底可能是什么东西。多谢！ |

as 1 sec for one scan with 101-200 m/z (100 m/z span): for every m/z unit (

m/z unit: only 1/100 second was used for accumulating a given m/z, and 99%

最近左上腹隐痛，不是刺痛，是那种钝钝的痛
也不是很痛，就是有时候感觉发堵，发胀
医生一开始以为是胃，但是去看了胃镜，没发现什么
后来照CT，发现在spleen内侧有一个大约一个立方英寸的mass报告上说，这个mass
partially calcified，没和spleen相连，不知道是什么东西好像CT上说contrast之后
这个东西的成像没有明显变化，但是也不能完全排除是恶性的。现在换了肿瘤科的医生
，推荐做PET scan。这个医生也说不知道是什么，有可能previous infection留下的东
西等等，只是猜测。
心中十分不安，所以上来问问。spleen内侧靠近心脏和adrena的位置应该是什么啊？我
自己想会不会是淋巴癌？PET scan准确率能有多少？我想直接做biopsy，但是医生说
biopsy太局部，有可能漏掉。。。
这个肿瘤科的医生只看了CT的报告，都没看CT图片，感觉都不怎么认真。。。
版上有没有人能帮忙看看CT图片的，给点意见好吗？
孩子就要出生了，这个消息就像晴天霹雳

PET scan做完了，那个mass没有亮
肿瘤科的医生说99%应该不是恶性的，至于是什么，还是不知道，连到哪里也不知道
怀疑有可能是长在adrenal gland上的瘤，也有可能是cyst，也有可能是gastric
diverticulum。。。
肿瘤科的医生认为，因为我能感觉得到这个mass并造成不适感，他建议切除，并把我
refer到了一个外科医生那里
明天就要去见外科医生了，心中还是比较忐忑的，毕竟还是不知道这个东西是什么，而
且对于接下来要怎么做，也没有概念。
大家能给点建议吗？我自己想，先要搞清楚这个东西是什么，连在哪里，etc
可能要先开个口子在体内成像？然后再决定怎么切？还是直接切呢？
这个东西的位置感觉挺隐蔽的，可以做微创手术吗？还是要开大些？
这种手术危险吗？死亡事故的概率大吗？如果正常的话，要休息多久呀？
