fluorescence), 你搞个屁啊。兄弟，那里面的动力学那个叫复杂啊。有个arthur
fontijn的，一辈子就是搞这类东西的。喜欢查文献的可以去查哈。人家刚毕业就有
nature。一晃多少年了，老头都没正真意义上搞出火焰动力学的应用。搞这个的在美国
不超过20个人。但是这个方向没有不知道这老头的。老头当时受了美国空军75万。结果
屁都没搞出来。你知道么，这不是简单的说加了引发剂就一定好的。复杂自由基反应那
个东西很黑暗的。当时一个著名的案例，就是汽油燃烧的调节。是加的四乙基铅。前后
2万种化合物的选择。外加前后做金属化合物的实验，死了上百人。fontijn组就有类似
的人。那哥们30出头。好不容易从他那毕业了。找了份烧垃圾的工作。结果，就半年，

ES细胞分为naive和prime两个stage
1)naive stage：ES colony在形态上看起来更“致密”，呈三维结构，对胰酶不敏感（

that the ES clones are targeted correctly. But i am not sure if the CreERT2
is functional or not, therefore, i want to detect the function in vitro ES

I only used the ERK inhibitor, and that was several years ago. The inhibitor
did improve the derivation efficiency. Having the p38 inhibitor in the
medium might help. You may also want to be sure that LIF is working well,
and i think this is very important.
The most tricky part to me is the patience. If you don't see any colony at
the first a few days, don't give up and just wait for a few more days.

是female mESC吗？是的话，先做两个实验：
1）H3K27me3 immunostaing：可能发现small dot in nucleus.
2)qRT-PCR测Xist的表达，或者用FISH。
证实的话，就说明出现X chormosome inactivation了。
几个问题：
3) mESC的培养条件？serum,还是2i/LIF?
1）knockdown的mESC是否仍保持未分化状态，还是开始分化了？如果未分化，那么很有
可能你发现了一个新的调节X reactivation/inactivation的因子，做深了，至少是
cell stem cell.如果细胞分化了，那么X chromosome上的genes表达下调不奇怪，有多
大新意取决于你的这个gene
2）看一下这个基因的功能，是否已有先关报道，与X chromosome reactivation有关。
是histone modifier, TF还是在X chromsome上的新lncRNA? |
