不是这个领域的人，结果就是这片文章很遗憾的忽略了一个很重要的工作，
在Lander的文章中，figure 2把CRISPR的发展划分成了三部分：
绿色：发现阶段，1993年一篇发现repeat的工作，和2003年的2篇文章
红色：2006年后2012年关于机制的
蓝色：2012年editing part
可见Lander也认为最初发现这个系统的阶段是很重要的；但正是这个阶段Lander忽略了
一个工作：在
2002年已经有一篇重要的计算生物学的文章：A DNA repair system specific for
thermophilic Archaea and bacteria predicted by genomic context analysis 发表
在Nucleic Acids Research。
这是一篇与1993年的文章的完全独立的工作，系统的报道了archaea和细菌中的DNA修复
系统，也就是后来的CRISPR系统（换了个名字）！并且这篇文中在CRISPR领域得到了广

我打算车后排放倒后可以放个床垫躺着， 比如amazon的 Car Mobile Cushion Air Bed

发帖数: 1210 | 4 Lander的文章确实有故意拔高Feng zhang的嫌疑，给了大段的煽情描述，不过Zhang在
Crispr用于genome editing方面，绝对是贡献最大的。
但是不管是不是拔高了Zhang，Doudna的贡献并没有被贬低。她绝对是个抢credit的好
手，整天的给自己打广告，恨不得抓住机会就说她是发现者，呵呵。Crispr这个系统的
基本生物学问题Doudna没有贡献，她只做了纯生化的工作。然而她的这篇唯一的重要文
章，还是Charpentier领头做出来的（doudna只是次要共通讯作者）。
crispr的发现过程和其生物学问题，zhang和doudna都没有任何贡献。现在问题的核心
就是，谁是最先用crispr做的genome editing？目前zhang和doudna的专利之争，也是
关于这个方面。目前的小道消息是，zhang绝对早于doudna和charpentier，有很清楚的
notebook为证据。据说这也是为什么Lander敢发表这篇文章的原因，因为他们已经确信
能够赢下这个专利之争。

我个人感觉：套用章良哥的一句话：21世纪是非传统模式生物的世纪。
crispr的发现和NGS的发展使得传统低等模式生物相比于其他生物的遗传清晰，操作方
便的优势变得非常的marginal，以后一定会看到更多更有意思的生物学被发现，而且很
有可能会直接促进我们对自身哺乳动物系统的认知和操作 |

Flag pole's shadow is on a different line most definitely not parallel to
the near edge of the moon lander's shadow. Which means, any line drawn on
the moon lander's shadow is not the true direction of the sun. Or, it means
the ground surrounding the lander is not flat, which would render any line
drawn on any shadow useless in determining orientation of the sun.

Delta。偶们先回到大温哥华。在河的南面最西面靠海，还有一个地区是划归于大温的
，就是Delta。有Tsawwassen，Ladner和 North Delta。从Richmond向南走99号公路，
通过一个隧道，就是Lander的领域，Lander给偶的印象也如同走廊一般，并没有太大的
特色，但是至少有两次，Richmond在下雨，偶们开着车走在９９号走着走着，雨没了，
再往后走最后还出了太阳。Lander的水域有不少floating House，感兴趣的朋友们可以
去看看，不过价格也不非，还没有地。Tsawwassen如同一个海角向南突出，是大温地区
和河谷地区降雨量最少的地方了，这一点很吸引偶，可惜那里的地价比Richmond还要高
不少,所以也没必要细看了。至于North Delta，简直就是和Surrey连在一起的。

连续五天的降水过后，Grand Teton雪太多，温度太低，没爬
Lander呆了两天，爬了Wild Iris和Sink Canyon。这个小镇在Yellowstone的东面两小
时车程的地方，镇中心的公园草坪上可以免费camping，有水有厕所没shower。Wild
Iris名字很好听，在Lander南面开车20分钟的地方。Limestone，主要的路线集中在
Main Wall。可惜Main Wall向南，夏天爬太热，我们只爬了OK Corral。路线都很短，
质量一般。因为有树荫人还挺多的。好多初学者，家庭在那边。爬惯了sand stone，
limestone的摩擦力比sand stone小，爬得我小心翼翼的。
第二天去了sink canyon，在Lander西南十几分钟的地方。比Wild Iris还热，我们在
Sink的main wall爬了几条线就走了。路线比较长，还是limestone。sink canyon得名

product a decidedly different shaped shadow than your straight box.

11.4号70mm IMAX场，提前接近一个月就订好了票，连基友都没有带就准备一个人
静静地观赏这场电影，免得到时候在回去的路上听人balabala说哪里不好看、又比不上
哪部电影之类的废话。好吧，其实是因为基友品味太差，只喜欢R.I.P.D这种类型的片
，实在提不起兴趣提前一个月订这种高质量电影的票。
今天最后提前半个小时就到了场，然后发现中间的位置居然已经被全占了，卧槽
，都是提前一个小时就来了么。最后只好选择坐在了最科学的观影位置，黄金分割线上
！
哦也，下面进入正题。。下面是没有具体情节剧透的观影感。
有几个基友问我好不好看，其实我有点不知道怎么回答。好看当然是好看，但这
又不是好看两个字能够涵盖的。把Interstellar总结为好看，我对自己的语言能力又开
始纠急。
首先，这不是一部想象力爆棚的电影，那部叫做Inception，但这是一部敢想的电
影。关于星际的电影，都恐怖异常。即便把标题告诉你，你也不知道，不敢想剧本可能

我们要不就是大早晨出发,带几套仪器,一个点一个点的架上,然后到下午
再一个点一个点的收回来,要不就是住在motel里,早出晚归,附近的测完了
就换个地方住motel.
记得去年刚刚发生地震后,我就主动请缨去观测---其实主要想
出去过过另外一种生活.landers那个地方因为92年的一个大地震一下子
闻名地学界,我们还以为那至少也是一个小镇子,应该有不少人居住呢.
按照地图找到的却是一片荒漠,路边有三两个小小的motel,开motel
的人还总喜欢养几只脏兮兮的小狗,或者是脏兮兮的马.我们继续开
着车转悠,希望发现点奇迹.果然,功夫不负有心人,我们又在一条
小土路边上发现了一个小邮局.邮局里还有人.我们象是见到
救星一样赶紧进去打听landers downtown在哪里.邮局的人很幽默,
告诉我们说,这就是啊,你没看到这里还有一个邮局吗?
由于那三两个小motel都住满了,这光秃秃的沙漠里一个小小的
邮局就是downtown了,我们苦笑不得的去打听附近还有什么地方有
motel.

，因为我们要质粒的时候还挺早的，那时候这些实验室自己的编号和后来他们放在
addgene上的编号不太一样。你去addgene上找landers lab的质粒就很容易能找到。不
过我们用下来觉得第一，他们cas9的质粒做成病毒后滴度太低，第二，这个质粒泄漏的
比较厉害，虽然号称是可诱导的，但是用flag染色还是能看见不少阳性的细胞。 |
