我的背景：某湾区独角兽hiring manager，过去几年中接触过各种级别几十个offer，
也在FLAG工作过，朋友众多，熟悉FLAG的情况。经常参加interview和offer decision

现在公认的序列是：DYKDDDDK
这个标签的好处是一来比较小，二来比较亲水，所以一般不影响所标记蛋白
的活性。至於是在N还是C端，要看你蛋白的实际情况。比如有些蛋白N端或者C端要
经过蛋白酶切除，这种情况下FLAG显然不能加到要被切除的一端去。当然你也可以
两端都试试，加在哪端活性好，就用那端好了。
如果是N-terminus Flag，ATG应该加在FLAG前面，之前还应该有Kozak sequence,
以便翻译正确。不用隔太多氨基酸，用一两个glycine之类旋转比较灵活的氨基酸
隔开就行了。
如果你想省事，可以找商业化的有FLAG载体，比如Stratagene的
pCMV-Tag系列,既有N端FLAG的载体，又有C端FLAG的载体。又如SIGMA公司的pFLAG-CMV
系列，也是既有N端又有C端FLAG载体。SIGMA公司还提供一种特别的p3XFLAG-CMV系列，
加有三个串联的FLAG。如果你的蛋白做western检测起来信号很弱，加三个或者更多

但是看到有人被这种文章误导，我老只好捏着鼻子找找抹黑之处来博尚粉以及欣赏尚的
同学一乐：
1. "唱功没有优势，音色都非常平庸"。
唱功没优势的评语不做置评，但说音色平庸就太扯淡了。音色这种东西属于各花入各眼
，对于某个歌手的声音，有人喜欢，有人厌恶都是正常的。尚的音色绝对算不上顶级，
但无论从哪个角度讲都算不上平庸。她一个在校大学生，从没有受过任何音乐专业训练
，甚至连简谱都不识的人，能拿超女冠军，如果音色平庸，就算她演唱技巧再好，演唱
感情再丰富，你觉得可能么？
2. "她从一开始到最后就反复强调自己是玩电子乐的"。
这个更是把flag混淆是非的能力展现到了极致。尚的确说过她是做电子音乐的，也不止
说过一遍，但节目开播以来，估计最多数次。到了flag嘴里，就变成了从一开始到最后
就反复强调自己是玩电子乐的。你说，这不是成心抹黑这是什么？
3. "她觉得要展现一些高端的东西"。
这又是典型的泼脏水逻辑。尚的确有表达过电子音乐比较前卫，担心不被观众接受的意

新手我最近苦于纠结一个关于IP实验的问题，特发此帖向大家求教。
我的目的蛋白A如果在肿瘤细胞（此蛋白已证实该细胞里正常水平不表达）里过度表达
会含有FLAG tag（通过adnovirus vector infection）。我用此蛋白来pull down 细胞
里的另外一个蛋白B。我用FLAG 抗体IP, Western blot 检测此两种蛋白A和B。结果在
正常对照的样品发现anti－Flag 抗体也能非特异性的结合蛋白B，即在blot也能检测到
B的条带。
请问如何消除抗体非特异性结合其他蛋白，还是我在那些方面需要改进？谢谢。 |

Flag.
用His6做，通过和Negative (untranfected cell)比对，还能找到表达的特异性条带。
用了Sigma的Flag M2 mouse做，保守估计几十条带。。。
不是说sigma的M2很好用吗，做了几次都这样。
大家能否推荐个好用点的Flag抗体，不介意flag-HRP conjugated的。

tag，还有一些是Calcium dependent，你看你用的抗体和你的FLAG tag是不是

，找了老半天都没找着（连四种颜色的也没找到），代码只好自己写了.
今天写 DNFP 四种颜色排序基本写对了，但是某些 test cases 不能通过。主要难度在
于当此颜色为第一颜色的时候应该怎么交换。研究了老半天，终于开窍了。原来是忘掉
了一个很重要不变量（invariant），就是 0 <= r <= w <= b < n （这里指的是原题
的三种颜色，r=red, g=green, b=blue，扩展至 k 种颜色的不变量为 0 <= mid[0] <=
mid[1] <= ... <= mid[k-1] < n）.
DNFP 四种颜色写对了之后，思路就清晰了，非常容易扩展至 k 种颜色。利用一个内循
环，来保持不变量 0 <= mid[0] <= mid[1] <= ... <= mid[k-1] < n. 这内循环似乎
很耗时，但少了这内循环，我又不知道怎样才能保持以上的不变量，望高手指教.

但是做为一个规模几百上千人的pre IPO公司CTO，从知识深度上讲，你不具备。从技能
广度上讲，你不够。
带过团队吗？没有，指导如何打交道吗？
我还是觉得如果你是个安安稳稳的码工，就在flag领高薪吧，过5-6年拿够公司的高薪
了，历练够了再
说海归创业。
最重要的，有些人带着侥幸心理，觉得回去忽悠一下就可以发财。哪有那么简单，国内
到这个程度的公司那都是精英，不说人品，只说能力也是国内万分之一。
还有，不要鄙视中国互联网，是，差远了。遥看5年前你的同学同事回国发展，很多都
比你现在强多了。公司不是被BAT并购发了一笔，就是做到了高职可以轻松投资移民美
国，你还在这里等绿。
要我说，机会还有2-3年，抓紧时间卡位去吧。趁硅谷还是一个高大上的光环

这是我参加的第一个 flag football season
学了很多东西，想来很多在校生是有机会玩的，我就介绍一下这个运动
据说flag football 的规则是全美统一的，我玩的是七人制八十砑
中线是四十砑，接着是二十砑，然后是goal line.
first down 并非往前十砑，而是 reach next line.
最长的first down 有二十呀，最短可能只有inches.
有touchdown, 没有field goal 和 PAT， 达振之后
有一砑一分，五砑两分，十砑三分 三种conversion options.
每人腰上束一布带，三条flag系于布带上，在两侧和后面。
布带被敌人拽掉即为down, 没有fumble, except airborne fumble.
有punt, 有return.
今年全校大约有八百个队报名，我们所在的grad/staff/faculty 大约
有二十只队。
现在讲讲我在的队，队名叫 BALL DEEP, 名字不错， 有十五个成员，

有耐心，请指点一下。:-p
makefile里面，经常看到用-D来specify flags，这是什么语法？我google "makefile
+ -D"找不到任何说明。
另外，好像通过CFLAGS能够define一些flags,在程序里面(c++)里面可以用（#ifdef
flags...),这又是什么道理呢？

发帖数: 89 | 49 新手我最近苦于纠结一个关于IP实验的问题，特发此帖向大家求教。
我的目的蛋白A如果在肿瘤细胞（此蛋白已证实该细胞里正常水平不表达）里过度表达
会含有FLAG tag（通过adenovirus vector infection）。我用此蛋白来pull down 细胞
里的另外一个蛋白B。我用FLAG 抗体IP, Western blot 检测此两种蛋白A和B。结果在
正常对照的样品发现anti－Flag 抗体也能非特异性的结合蛋白B，即在blot也能检测到
B的条带。
请问如何消除抗体非特异性结合其他蛋白，还是我在那些方面需要改进？谢谢。 |

发帖数: 354 | 50 做了个flag-tag fusion protein，在293 transient transfection, 用蛋白本身的抗
体检测是一大坨，q-pcr 表达也是有几十倍的升高。
但是就是用M2 beads IP不下来或者极少。
然后用flag 抗体检测，居然多次检测不到。最后是在疯狂一下地翻了10倍的二抗量 膜
都要压爆了才压出来条带正确的带，大小正确，量也与基因本身的抗体显示的量对应。
这在本实验室在293瞬转检测中似乎从来没有遇到过。
难道是flag跟我的蛋白在一起就难以检测？ 甚至在denature gel中？
那在nature状态下更难结合？所以IP失败？
