过 RNA 调控基因表达的机制, 它是指在真核细胞中引入双链 RNA分子从而导致具有序
列同源性的基因产生特异性基因沉默 (gene silencing)的现象 。由于 RNA干扰可以阻
断特定的基因表达, 具有超越疫苗和抗病毒药物的诸多优点, 因此在阐述基因功能以及
蛋白质相互作用等方面, 表现出诱人的前景。近年来随着分子生物学的发展, RNAi 的
机制也不断被阐明, 同时国内外研究也证实 RNAi 技术可以为动物繁殖过程中所涉及的

如果室温24左右的话,L1虫子2天过后基本就是刚young adult,开始产卵但不是很多得
水平,孵化出来需要大概10小时样子,所以如果是野生型,周三早上应该看不到F1得
larva,P0得当然还在。
第二题:
dpy 虫子一般是表皮和结缔组织有问题吧.collagen突变什么得。但是也有影响神经得
。对生殖应该不影响吧。 |

2.没做过遗传筛选,不知道题目中的F1到底是哪一代。推测大的那只虫子的表型可能是
母源调控的(它妈妈赋予的),而虫子自身的基因型可能是纯合野生型,所以当它作为
母亲时,后代没有表型。纯属猜测。 |

1:看一下下面网页的life cycle,自己算一下从L1到adult需要多少时间,然后需要多
少时间从egg孵化到larvae

2:如果有normal head,那么应该epithelia没有影响。如果能够有后代,那么说明
muscle没有问题,reproductive tissue应该也没有问题。剩下两个你自己继续排除。
另外你可以到 www.wormbase.org 去查查 dpy mutant一般和哪些基因有关,这些基
因的功能分别是什么
3:在F1就分离到mutant,说明mutation is dominant。没有看到遗传到这个性状的后
代,说明很可能这个mutation同时也是 dominant sterile或者 dominant lethal。之
所以在F1能看到,多半是maternally rescued by P0

at NIH RePORTER. I searched for active projects/R01 equivalents at NIMH,
NINDS, NIAAA and NIDA. Turned up about 400 projects with the key word
elegans, drosophila or zebrafish. Another 1,500 with mouse. Very
interesting sociology of basic research. I'm glad that you motivated me to
do the search!

动物应该是很难的吧?换言之,我们应该是缺少这方面数据的。而仅有的少数数据应该
也是驯养的吧?这就好比我们在实验室做c elegans/drosophila一样
当然了,如果驯养动物的同性恋比例真的比野生的高很多,比如你说的这个10%,我感
觉真的是很高的比例了,那还或许真是这个挑选驯化过程会对性取向有影响。这是个很
有趣的观察角度。
其实我觉得要从进化的角度解释同性恋,首先一个很根本的话题就是:请从进化的角度
来解释“性”,以及“性”和“生育”的关系。
要讨论这个问题,先要说明到底什么是“同性恋”。这听起来非常愚蠢,但坦白说我觉
得很多人根本不知道何为“同性恋”,包括很多pro-gay的亲友甚至同性恋自己也不一
定很清楚。比如我遇到很多liberal派的人,他们说支持同性恋,就是因为只要是真爱
,所以就支持。我当然很感激这样的人,但坦白说他们不知道什么是gay。只要是真爱

我的mutant实际上还有很多noise,原因是我的线虫体外回交成功率极低,这也是为什么
而且,我研究的线虫基因组还没有发表,所以也没有common variants数据库了,我这
个搞非模式生物的也只能流口水
所谓的wildtype A和B,是同一个isolate里发现的,理论上来说genome的variation很
小,测序数据也证明了这点
其中一个问题是,两个wildtype的差异基因很可能是控制上游表达的,而mutant中的
candidates基本都靠近下游。这是通过研究c elegans的ortholog 得出的结论。但不是
很肯定,因为线虫间的差异很大。
另外,我手上还有两个wildtype在stage L2的transcriptome,这个stage是所谓
phenotype的决定期。通过分析发现不到30个基因差异表达。有没有什么方法可以把
genome和transcriptome放在一起做个cross analysis的?
你上一个回复中提到的structure variation,是指rna的还是蛋白的?是怎么做呢?如
