用4-OHT做gene inducible deletion5天，然后wash out 4-OHT以后，E14.5的mutant
细胞相对E14.5的control cell line proliferation降低，但是E13,5 的mutant cell
相对control cell line的proliferation居然升高了 （我每个时期的细胞系都有2个
control 2个mutant），一头雾水呀。 |

我一般都是seed 1x10^5 cells/6 well. 细胞生长的也不是很dense，而且我重复了一
次，结果是一样的。搞得结果很难解释。我现在就是想是不是这个OHT对我的细胞有影
响，还是我研究的这个gene 对 OHT treatment 有response。
其实这个gene我们propose和stem cell 的分裂和分化有关，一个naive的想法就是E13.
5的primary细胞是不是保留了更多的stem cell identity？ |

我们是用小鼠胚胎的软骨和骨头染色的protocol。染软骨的话只用Alcian Blue即可。
最后胚胎用BABB处理就会变得透明。BABB危险，而且会腐蚀显微镜，使用时要小心。很
多动物的胚胎都可以用类似的方法染骨头（红色）和软骨（蓝色）：鼠，龟，蛇，鸡，
鱼，蜥蜴，变色龙，蝙蝠。。。而且都很漂亮！

发帖数: 236 | 31 我在德国待了7年，在美国2年，给lz一些参考吧。我是理工科的，对医学方面不了解，
只说我知道的情况。德国教授职位很少，如果没给你教授头衔那就不要想当然说自己的
职位相当于教授了。但是德国的永久职位也很少，不知道医学院啊，如果是自然科学这
类的话，拿到永久职位是很不错的。很多研究所 大学招博士后不会让你干满五年，合
同都是短期的，过几年就让你走人，不然干满五年就要给你永久合同了(几年前是这样
，最近有没有新规定不知道哈)。有些永久职位像lz这种，可以自己招人，有自己支配
的经费，是很不错的。因为德国教授位置比较少，lz这种位置就很好了。有些热爱科研
的人还就喜欢这种位置，不需要像教授一样到处飞 拉赞助 教课 维持人际网等等琐事
，可以潜心搞研究。
消费，美国各个州不一样，同样德国也类似。我在德国两个大城市住过，其中一个德国
消费最高，另一个也能排进前三，目前住的地方消费在全美可能排不上号但也挺高的，
这边超市食物比德国平价连锁超市贵很多，当然德国也有一些贵的超市和这边价格差不
多，但是起码你可以选择。房租这边比德国贵。汽油和餐馆比德国便宜。

位置的，不都是博士后吗？只是说法不同，就像在美国可以叫博士后，可以叫research
associate, research associate professor 其实都没有区别。
即使E13， 熬年头也有5万9.就工资而言不见得比教授低，教授的好处是
Krankenversicherung: -
Pflegeversicherung: -
Rentenversicherung: -
Arbeitslosenvers.: -
这些都是雇主支付的，所以他们拿到手的还是高一些。 |

发帖数: 1704 | 35 博后工资按TVL-E13最低级别也是4万出头了，如果是德国本土的博士（LZ就是），读博
期间算工龄的，那么就至少是3级了，大约4万8。没听说过博后不给全职的，除非part
time job。至于税后拦腰砍，呵呵，单身的话税后大概是税前的60%，但是医保养老保
险失业保险等等全在里面了，来年还能申请退税。
基本上博后可以直接申请欧盟蓝卡，最多5年可以拿到欧盟长居或者德国长居（绿卡）
，如果是本土博士甚至更快，很多人毕业之后一年就拿了。
所以除非特殊情况，很难有动力往美国折腾了。 |

第二匹克：XOM
这是美国能源财团的旗舰股。著名的能源反指，Bank of Montreal已经发布了明年原油
20的警告；所以童鞋们尽管放心大胆地捞吧。目前P/E16，红利3.72%比任何CD都强。
第三匹克：ADM
大湿非常同意版上对El nino现象的分析，和美国粮食趁机涨价的计划。民以食为天，
星球大战看完了也得吃饭；没了粮食，咱们这点原力撑不鸟几天。ADM是全球四大粮商
之一，巴菲特曾持有此股；所以公司没问题。

category. 1.5 years sounds long. If it is already outside your normal

I-140 on E13 category @ TSC shows 4 months. I asked the lawyer and he said
it is normal. They requested for fast decision and TSC respond that it is
within a supervisor review. |
