Cells" from invitrogen. I think TOP10 and DH5α are similar, except the
former having better efficiency.

it seems to me that the difference exists with the F' factor.
i think DH5a is typically used for white-blue screening by
alpha-complement and also a common host for plasmids. JM109,
according to the comment, supports growth of vectors carrying
amber mutation and will modify but not restrict transfected

Purpose: Insert 9kb mouse sequence into vector pPNT6.
Vector pPNT6: Its map is at http://www.addgene.org/pgvec1?
f=c&identifier=11072&atqx=pnt6&cmd=findpl . I have inserted 0.9kb DNA in
this vector at XhoI/EcorI
double sites. I should have no problem in cloning short fragment.
