If I want to loop through iterator and delete some elements on condition, I
have to do it:
1. Loop through iterator and find all elements satisfy condition, save them
in a collection.
2. Delete temp collection elements from original collection.
I have to do two steps because iterator can't be broken during loop. Is
there a better/simpler way to do this? I have no limitation on the data
structure.

就像你说的，snp有correlation，他们之间不是完全的independent。snpA的表达可以
影响到snpB的表达，特别是他们位置上很近。
conditinal probablity的公式：
p(A|B)=p(A交B）／p（A）.
p(B｜A）＝p(A交B）／p（A）。
所以，condition A 观察 B 的变化， 和 condition B 观察 A 的变化是不同的东西。

1. CreERT2 (就是第二代的CreER). Cre driven by tissue specific promoter.
我研究骨头。打了tamoxifen （我用oral gavage)10天后就有phenotype。但是
tamoxifen有些副作用，比如增加bone mass.所以需要optimize dosage.另外我用打了
tamoxifende wt mice 作为control,这样可以排除tamoxifen effect.
2. 我用过Tet-Off system. 需要三只老鼠：tissue specific-tTA; TetO-cre,加上你
的conditional 老鼠。喂老鼠doxycyline-containing water or food 直到你想要表达
基因的时候。

mouse gene function". 这里的ESC叫什么knockout first的conditional,
貌似本身就是KO，然后Flp一下变成wt, 最后再Cre一刀变成KO，我的
理解应该没错吧？
问题是，这种东西应该是heterozygous的吧？因为一开始就是KO，如果
直接打老鼠，还没有flp之前，如果是致死或者发育基因的话，应该不会
影响打老鼠吧？还是打老鼠之前就要flp一下？

PBS就说PBS，大家都懂。不要认为就是生理条件。很明显不是。比如PBS里面没有Ca2+
。在任何做电生理的环境中，都不会说PBS是physiological condition的。
我猜你遇到的情况是PBS是对照，另一组是PBS加某药。那很简单，你就老老实实说PBS
是对照组就行了。 |

主要是为了研究medium的细胞因子，生长因子。 比较了几个不同的protocol，有几个
不同点
1. RPMI Vs. DMEM, 有的protocol不推荐加抗生素，有的在收集CM前12小时换成no-
serum medium,有的一直在用regular full medium. 不知道血清里的生长因子细胞因子
会不会背景太强（用之前是heat inactivated过的）。
2. 如果为了鉴定一些特定因子x，我看有一些protocol用抗体在收集的培养基里加入x
的抗体。 和lab的人讨论过，他们说这种免疫中和的实验不是很好做。。。（也没说出
个所以然，只是说抗体的affinity可能很低balabala）。 然后有建议说用shRNA KO,
我试过c26 gfp 转染，感觉效率很低，当然lentiviral可能好很多。
第一回做这个protocol，希望有经验的大牛能够指点下。谢谢。 |
