From the papers I have read and the experience my labmates have, I learned
that they all use Concanavalin A
(conA) to recognize leukostasis and use CD45 to confirm the counting.
But I don't understand why use conA as the particular lectin to do this
experiment?
Is it because the leukocytes express abundant mannosyl or glucosyl sugar
groups that only conA can bind to?

目的 流感是一种由流感病毒感染引起的急性呼吸道传染病,具有发病快、传染性强等特
点,曾多次造成世界范围的大流行,严重影响人们的日常生活和身体健康。对于流感的防
治,目前还没有安全性和有效性都较理想的方法。大蒜油有防治冠心病、酒精性脂肪肝
、杀菌、抗癌、杀虫等多种功效,其抗巨细胞病毒、肠道病毒、鼠肝炎病毒、生殖器疱
疹的研究已经有了初步进展,但大蒜油抗流感病毒作用的研究还未见报导。本研究通过
体外培养的细胞和动物模型,对大蒜油的抗流感病毒作用进行了研究,并对其机制进行了
初步的探讨。为大蒜油用于流感的防治提供了科学依据。 方法 1、流感病毒的增殖：
将0.2mL人季节性H1N1流感病毒(以下简称流感病毒)接种于10日龄的鸡胚尿囊腔内,于34
℃孵育箱里孵育44-48h,每日照检,收获前放于4℃冰箱6h。无菌吸取尿囊液,接种于下一
代鸡胚,方法同上,直至尿囊液中流感病毒的血凝滴度>1：128。然后将符合条件的尿囊

（十八）凝集素层析
我们用去垢剂溶解了膜蛋白之后，紧跟着的一步就是用Wheat Germ
Aglutinin(WGA) agarose来部分纯化insulin receptor。WGA是凝集素
(Lectin）中的一种，而Lectin指的是可以与糖基结合的蛋白质。现在常用
来做蛋白质纯化的凝集素绝大部分是从植物中提取出来的。绝大部分膜蛋
白的extracellular domain和分泌蛋白都有N-linked glycan 的糖基修饰。
根据这个性质，用固定有适当的凝集素的beads，比如WGA agarose或
者ConA agarose可以部分提纯膜蛋白。注意，只能是部分提纯，凝集素
层析并不能一步登天，让蛋白质比活力一下子提高上千倍，至多几十倍就
已经很不错了。但尽管如此，凝集素层析是一种很好的办法，因为buffer
条件非常温和，洗脱液是加了糖的buffer，透析什么的也方便，非常非常
适合于和其他层析方法偶连起来用。
纯化膜蛋白用的最主要的两种凝集素是WGA和ConA。谈到这里，
