该相信。
做过一些迁移的实验，多说两句。
1 transwell assay: 细胞状态很重要。你如果是贴壁细胞，trypsinization时间的些
微差别，离心重悬吊强度就有影响。另外我非常怀疑公司的产品是否能够做到不同孔的
膜都做得一样。
2 in vitro wounding assay：就是贴壁细胞划线，然后在特定时间看迁移到细胞所覆
盖的面积。此assay对medium的成分非常灵敏，要严格按照别人的protocol来做。换个
medium结果竟然会相反！你说该信哪个？此assay的定量分析非常耗时。
3 needle assay：就是一个小needle尖头释放chemoattractant，然后用软件实时记录
每个细胞的迁移，运算得到迁移速度和角度。模式生物Dictyostelium做这个assay的时
候需要先polarize才能迁移。很多文章说某个突变体影响力迁移速度。问题是很多突变
体本身它没法被polarized。你说究竟是它的极化能力受到影响还是本身的迁移受到影

the ligand that can bind to your chemotaxis receptor, which cause receptor
internalization and desensitization. Incubate them for 15-30min in medium
without serum or starve them overnight will resensize the migration and
increase receptor expression. If you are sure your culture medium does not
contain the ligand, you do not need to starve them. |

That's not my project anyway. We are not a neuro lab and my boss is doing it
herself.
The repellent is a social signal. The screen was done on genes known to be
involved in chemotaxis/olfaction in
this animal. No GPCR is tested because there are too many. However key
components of the GPCR pathway are involved.
I suggest my boss to identify the components of the repellent using HPLC and
do some calcium imaging instead of using laser ablation only. My boss says
