话，cell sorting或Puro selection不是可以解决吗？
我只觉得两种情况下需要virus delivery system--
1。Pool all cells, no need to clone cell line。这个siRNA里经常的问题。但
siRNA有时间问题，transfection后不能等太长。而cas9是permanent deletion，又不
能保证transfected cells一定有genome editing发生，这和siRNA显然不一样。
2。deliver a library of guide-sequence-cas9-vector。这个我一直很感兴趣。最近
的两篇文章，一个library是针对18000个基因，另一个是73000个基因。这个对找一个

2. use inverse transfer function to synthesize a 'high pass' filter;

Mt. Adams位于美国西北的Washington州境内，南边距离美国第三大河Columbia river仅
30英里左右，是Cascade最东边的一座火山。Adams海拔12276尺，是Washington州第二高
峰， Cascade 第三高峰。和周围的几座火山（Mt Rainier，Mt. St. Helens, Mt. Hoo
d）相比， Adams沉寂得多，在相当很长的时间内处于休眠状态，最近的一次火山活动，
发生在1000年以前。现在的山体形状，主要形成于一两万年以前， 后来的小规模爆发，
以及日照，暴风雪，和冰川的侵蚀，对山的主体形状并未产生本质性的影响。
Mt. Adams首次被白人发现是1806年4月2日，探险家Lewis和Clark在他们的西行探险过程

了一下deling,
他爬的都是长的trail, 正在找一些短的 day hiking trail. 最理想的情况就是在公园
里住下来，
然后每天走走trail, 不过老觉得north cascade比较荒，一个人好像有点不安全。 |

东北边从sun rise附近, 可以到glacier basin, 也能算个day hike吧
西边也有一些, 现在雪还太多
north cascade可以走cascade pass那条trail, 也是个day hike, 其他的就在那几个湖
附近转转.
