处理的时候加CQ, 如果cq能够增强你的处理引起的LC3II 增加，那就是诱导autophagy,
如果没什么变化，那就可能是LC3II的降解受抑制导致LC3II增加，就可能是autophagy
flux blockage：fusion of autophagosome and lysosome is inhibited。p62是
autophagy的特异性受体，自噬增加会导致p62降解增加，减少p62蛋白量；flux受抑制
则会降低p62的降解，增加p62的蛋白量，所以你的情况看着像是flux blockage。 另外
，检测的时间点很关键的，没有记错的话饥饿2小时的时候ＬC3II 上升，4小时之后则
降低。 推荐：Guidelines for the use and interpretation of assays for
monitoring autophagy |

upregulation, induced autophagy, 也有很多paper说抑制了autophagy看到p62的
accumulation。这个p62上调到底是说induced还是suppressed autophagy。越看越糊涂

The title is certainly overstated. There are many cavities in this work, to
name a few:
1) CCI -> mTOR inhibition >> autophagy: may or may not related with
autophagy.
2) CCI suppresses necrosis?: not well-defined.
3) Is downregulation of RIP3 a cause or consequence of attenuated cell death
? Not answered.
4) How does CCI regulate the RIP1/RIP3 levels?
... Plus, the quality of the data is not that great.

dogma approaches only represent the ideas of a few labs including Klinosky
and Levine. Autophay is such a sophisticated machinery that's responsive to
a diverse array of stimuli. I hardly think p62 is a universal substrate for
all types of selective autophagy process. Granted that not one single
approach could be exclusively relied on, a combination of several key
experiment would triumph over p62 degradation. |

2：还是定量问题：看到说autophagosome大小在0.5－1.5uM，照片上的很多小点是小于
这一值的，是不是只数大于0.5uM的？
3： 怎么解决heterogeneity的问题。看autophagy文章error bar都很小。怎么培养细
胞才能减少对照细胞的autophagy？ 同样的问题在starve之后也存在。
4：细胞在EBSS里starve6小时，也没看见GFP puncta明显增加，是不是有什么没有注意
到的问题？还是在这个荧光定量的实验中，大家都是默认加inhibitor的比如bafA1，即
使实验方法中没有明确说明？

实在不行就看WB的LC-3B cleavage
荧光定量的实验应该是Semi-quanitive
inhibitor有很多,对autophagy不同阶段有抑制.所以要选好.
Acridine Orange荧光染色,可以用
micro-palte reader(定量)
flow cytometry(定量)
荧光micorscope (image)
另外knockdown Atg7/5等表达,在看autophagy在同样处理下发不发生 |

不过那是极少数的细胞，而且数目也并不多，在我看来应该是正常情况下的autophagy
不过，处理后的细胞很容易看出区别

来共同支持autophagosome的形成：
1. GFP-LC3转化后可以用来看puncta formation
2. 再有MDC assay有时候也还可以用，但比较tricky
3. 最有说服力的应该是TEM了吧，电镜下可以看到autophagosome或者autolysosome
俺不是做autophagy的，但一个小课题涉及到一些，说错了轻拍啊，呵呵，等牛牛们来
回答好了。 |
