IC50在100～200nM。但是用这个AF488标记的antagonist染色target高表达的细胞系，
在flow cytometry上面看不见那个波峰向右边移动，请问大侠可能的原因是什么？谢谢

发帖数: 884 | 5 最近做了个实验, 关于乙酰胆碱接受酶(acetylcholine receptor, AChR)抑制剂（
antagonists)的药物筛选。
我用的cell-based assay, 利用钙离子流量来监控AChR, 就是add agonist (nicotine)
to trigger Ca2+ release of the cells. If treat cells with nAChR antagonist
first, then the nicotine can not trigger Ca2+ release.
AChR主要分为两种，一种以 nicotine 作为 agonist, 叫做 nAChR；另一种以
muscarine 作为 agonist,叫做 mAChR.
我用这个cell assay试了几种药, 发现 mAChR antagonists 也可以抑制 nicotine 的
作用。我的问题就是，一般的 mAChR antagonists 可不可以当作 nAChR antagonists
来用？它们的作用是不是互通的？

ug 呢，还是指针对一个 signaling event，还是针对一种 bio-function？
我过去一直理解为，能跟某种 ligand 的 receptor binding 的东西，如果跟这种 li
gand 引起的作用相同，并且作用的程度类似或者更高的，叫 agonist；如果 binding
但不引起后续的 signaling events 的，也就是说，block 了 receptor 的，甚至引起
相反的作用的，叫做 antagonist；能引起类似的效果，但是不如 ligand 有效的，叫做
semi-agonist，又因为这种 semi-agonist drug 和 ligand 竞争在 receptor 上的 b
inding site，如果 ligand 过量而 receptor 不足时，或者这种 drug 的 affinity 高
到严重阻碍 ligand/receptor binding 时，这种竞争就会导致 signal 减弱，所以这时

观遗传学」（epigenetics）解释同性恋原因。表观遗传学主要研究环境如何改变基因
表现（gene expression），据指出，当胚胎在母体发育初期，能影响基因表现的「外
延标记」（epi-marks），会向与性别相关的基因发出指令，保护胎儿免受环境影响，
像避免胎儿免受过多睾丸素影响，这足以影响胎儿生殖器官发展、性取向和性别认同。
一般而言，外延标记不会遗传下一代，否则会以父传女、母传子的方式遗传下来，
令女儿较男性化、儿子较女性化，特别强势的外延标记就会导致同性恋倾向。但这理论
仍须研究确定。

戏剧。戏剧就是以一种故事的方式让你以代入的方式去感受历史，而不是像看纪录片一
样去理性认识历史。所以归根到底，你需要一个有力的故事。
有一本书叫叫，里边总结了戏剧story line发展的最基本
因素，就是character 和 antagonist之间的矛盾的产生，解决，新矛盾的产生（或者
旧矛盾的生级），这是推动戏剧发展的核心动力。这个模式用网络语言简而化之，就是
生级打怪，最后大boss出现，就是一般戏剧结尾高潮show down的时间。《狼牙榜》和
《越狱》都是这个基本范式的样本，所以尽管《狼牙榜》有很多小问题，它的框架是经
典模式。
值得指出的是，这个antagonist不一定是人，也可以是某种情势或者悬念。是上述基本
范式的衍生模式。而且不是每部戏剧都有一个从一而终的冲突线，这时就需要不断的埋
钩，定期起线。比如《知青》就属于这种，从开始的奇勇和小地瓜的私人恩怨，解决后
向前演进，而后麦收出现新的挑战，再然后出现禁书的问题，等等。

case, Zofran usually does not cause EPS. It was a serotonin agent( which
very rare causes EPS). Reglan/compazine are D2 antagonist.
If you do not consider Seizure as a differential diagnosis, your
consultation is incomplete. D2 antagonist lowers seizure threshould and
ocular movement could be aura or non-convulsive event(not necessorily affect
MS). It is very possible when you just leave the OB/GYN after you tell

青蒿素就是一个典型的例子， 一个中药， 几千年前没准就知道对Malaria有效， 但是
如果不是提纯出有效成分， 估计非洲人也不会去喝那些黑乎乎的汤水的。
青蒿素本身很不稳定，尤其怕热， 在形成过程中收外界气候土壤影响非常严重， 并不
是所有地上长的青蒿都会有青蒿素成分在里面的。 这个例子肯定也适用于其他的中药。

这也注定了只有提纯出有效成分（西医之路)才是中药的出路。
可怕的是, 今天的中药发展出来的所谓科学方法就是往里面加西药而已
比如什么中药可以治疗失眠， 其实里面加了个老掉牙的西药，某个Histamine
antagonist；
什么中药可以治疗高血压的，其实里面加了某个老掉牙的calcium channel blocker，
或者
adrenergic receptor antagonist。

一样。 individualized medicine对综合征这样的疾病尤其重要， 病灶在大脑，但具
体部位和发病机理千差万别而且不能明确诊断。 我一般会根据病人的具体症状尝试一
下药物治疗， 无效只能用Botox，当然两者可以同时使用。
药物类dopamine agnoist或者antagonist可能有效， 其中antagonist一般有效，但副
作用是TD，有人说dopamine antagonis本身就可以引起meige所以没有经验不要随便使
用。 AEDs也可以有效，而最常用的是benzodiazepine，副作用是limitation。
Clozapine是一些专家推荐的，没有经验不要用。 |

protocol，但是因为身体没反应最后取消了，医生说下一个cycle 让我用micro-flare
protocol。我自己在网上查了一下，好像说micro－flare protocol 和estrodial
priming protocol是针对我这样的poor responder 的，所以如果你对用药反应不错的
话，用antagonist protocol应该也可以。
