且顺利的拿到了Immigration Visa。打算两周之后去美国做immigration landing。
但是今天早上收到了USCIS的邮件，说是Department of State要求USCIS重审我的Case
。。
我所有的电话都打遍了（律师 USCIS NVC 大使馆）都说不知道，让我安心等待180天再
说。。
（1）我说我的Immigration Visa还有一个月就过期了，我还怎么等180天？
（2）如果我明天就去landing，边境是不让我进吗？我的VISA在手里面，也没有人说
VISA被revoke了
我也是傻眼了。。。请问大家有没有遇到过类似的事情啊？？？ 呜呜

including 25+ people for golf, other activities: hiking group finished 20
miles 3 Bond hiking, Salem Lighthouse Bike Tour (this tour is very good and
we will bikeit at least one more time), North End gathering, and Newport
city biking. Thank Kady, Catherine, David, Ning and Amy for organizing
these activities. You guys did a great job! |

表现为：
ant agent虽然显示说从手表上接收数据，并且上传到connect成功了，
但是在connect的列表中看不到这个活动，然后用connect的upload功能上传时，一直停
留在uploading状态。
随便新建两个activity，都能上传成功，估计不是硬件问题也不是软件问题，而是这个
activity的问题。
解决的办法：
放狗garmin connect uploading forever，看到有人提到fit文件修复的字眼，
然后继续放狗garmin fit fix，找到一个软件，要安装要license，还找到一个网站：http://garmin.kiesewetter.nl/
试了网站的，上传fit文件（C:
UsersucolAppDataRoamingGarminDevicesxxxxxxxxxActivities），然后修复后下载fit
文件，再去connect网站上手动upload修复好的fit文件就成了。

今天activate给verizon打电话，说必须要sim card才能activate，说是任何verizon的
iPhone5
版本的sim card都可以activate。
因为这些customer service经常信口开河，所以上来问问高人，是不是这样？能用别的
carrier的iPhone5的卡激活吗？

We use same inducer for all the experiments.
P should have same function with respect to X1-R2 and X2-R2. Once bound with
X1 or X2, both R1 and R2 should block the transcription of eGFP.
R1 and R2 recognize different inducer. The inducer for R2 is a universial

主要疑问是关于dCas9-gRNA binding site
文献上说法一般是在transcription start site （TSS）上游
激活特定基因的 会在TSS上游挑选8-10个位点 逐个测试挑个好的
genome-scale的话 一般就是每个位点挑3-5个位点
目前我们打算是做target pathway level activation （20-30个基因） 如果每个位点
都要搞个5-10个 工作量挺大的 又不能像genome-scale那样把所有DNA混起来
问题1.貌似TSS不是很清楚，有些基因有多个TSS，有这方面的数据库不？最好包括酵母
、老鼠以及人的
问题2. binding site有没好的软件可以预测？做knock-out的有，貌似做activation没
见到过？有啥推荐的不？ |

research article的intro部分直接一笔带过说CD28对T cell activation很重要。但是
仔细读文献发现CD28 CD4 T cell activation的重要性很清楚，但是对 CD8+ T cell
activation的重要性有争议，有正面有反面的证据。反面证据之一是CD28 KO mice对
LCMV有正常的CTL response，也可以reject allograft。希望专家出来解释一下现在主

回国两个月，回来后手机不能用。登录帐号发现电话被de-activated.跟客户服务联系
，说因为non payment.帐号里面的钱也没有。
客户服务说：只能重新activate,但是不能用原来的号码，必须是新的。还说：port
phone number只能从别的provider那里转过来。
手机比较旧，HTC ADR6300, 用的是verizon网络，pageplus pre-paid.
有没有别的verizon pre-paid/post-paid,价格也不贵的服务商？想用原来的号码。
