防系统研发工作已经接近尾声,将于在今年装舰进行测试。该系统是“铠甲”-S1型近
防系统的海基版本,在性能上获得了很大的提升。
一般认为,最新的“铠甲”-ME是“铠甲”-S1型近防系统和“卡什坦”近防系统的
混血,兼具两者的长处。该系统配备了“铠甲”系统的1RS2-1E型相控阵雷达,同时也
安装有光电/红外目标获取和识别系统。
“铠甲”-ME型近防系统在俄罗斯海军的大小舰艇上都可配备,比如“库兹涅佐夫
”号航母、“基洛夫”级巡洋舰和“卡尔库特”级轻型护卫舰都能装上。该系统配有两
门GSh-6-30K/AO-18KD型30毫米加特林炮,具备非常强大的瞬时火力。
单门GSh-6-30K/AO-18KD型30毫米加特林炮的射速为4000~6000发/分,而两门炮加
起来射速可以达到1.2万发/分。相比之下,早期的“铠甲”-S1系统仅仅装两门单管的
30毫米机关炮,单炮射速仅有2500发/分。因此,仅仅火炮的射速就提升了很多。

I am doing purification of membrane proteins to get enough concentration for
NMR study, the protein was labeled by his-tag
Firstly, the protein was expressed by E. coli, lysed by several enzymes,
agents and detergent(empigen) to solublize everything, then it was purified
by Ni-NTA column, the protein shows at the right MW(about 18KD) at commassie
blue stained gel.
However, the problem is, after the second purification---ion exchange on

or SUMO
当然,也不能排除磷酸化 |

sorry for not mentioning...and the 23KD and 18KD are also in the flowthrough
( but so weak compared to the peak I collected), it's possible that the
protein was trapped, but the amount of proteins seems the same to me,
although we didn't quantify... haha, thank you very much for answering |

f*******d 发帖数: 92 | 6 Histone 是很容易做的,要sonicate cell lysates, 确保细胞核裂解,RIPA buffer绝
对没问题。如果没有的话,1% SDS + heating也很容易看到,如果还看不到的话,用
8M UREA+RIPA, 这样的话,即使在胶上都能看到很强的band. 此外这个protein特别小
,好像15还是18kD, 所以胶别跑过了。 |
